Cholic acid is a bile acid, used in food as an emulsifier. JECFA's specification describes colourless plates or a white crystalline powder of not less than 98% purity.

Where it is and isn’t permitted

Status of Cholic acid by jurisdiction. Every jurisdiction this site covers is listed, including those where the substance does not appear.

Australia and New ZealandFood Standards Australia New ZealandNo listed food uses foundThis site has not matched this substance to any food use on this jurisdiction’s list of authorised food additives. A food additive may be used only in foods that list names. Flavourings, enzymes, processing aids and added nutrients are regulated separately and do not appear on it.
CanadaHealth CanadaNo listing matchedHealth Canada’s lists name additives without E or INS numbers, so this site matches them by exact name. A substance listed there under a name this site does not recognise has no row here.

Existing additives, natural flavouring agents and ordinary foods used as additives are permitted outside that list and are not imported, so an empty row does not show that Japan does not permit the substance.

What FDA’s inventory records

This page holds no listing for Cholic acid in the parts of 21 CFR this site reads. FDA’s Substances Added to Food inventory does carry it, as CHOLIC ACID, pointing at no regulation at all.

Technical effects FDA records: ph control agent.

The inventory is an information resource: a row means FDA holds information about the substance’s use in food, not that the substance is listed for that use. It is shown apart from the permissions above and is not counted as a US permission anywhere on this site.

What it is made from

Cholic acid is the principal bile acid of mammals and is obtained commercially from the bile of slaughtered cattle and sheep. Anyone avoiding animal-derived ingredients should treat it as one.

How JECFA defines it

The international specification for identity and purity, prepared by the Joint FAO/WHO Expert Committee on Food Additives and published by FAO. It is a different document from any above, written by a different body and revised on its own cycle; it is advisory, and a country adopts it or does not.

INS 1000 Cholic Acid

Synonyms: INS No. 1000

Assay, identification and purity criteria

Description and assay

Specification history
Prepared at the 17th JECFA (1973), published in FNP 4 (1978) and in FNP 52 (1992). Metals and arsenic specifications revised at the 55th JECFA (2000). An ADI of 0-1.25 mg/kg bw was established at the 17th JECFA (1973)
Chemical names
Cholic acid, 3alpha,7alpha,12alpha-trihydroxy-5ß-cholan-24-oic acid, 3,7,12-trihydroxycholanic acid
C.A.S. number
81-25-4
Chemical formula
C 24 H 40 O 5
Formula weight
408.58
Assay
Not less than 98% after drying
DESCRIPTION
Colourless plates or a white crystalline powder
FUNCTIONAL USES
Emulsifier
METHOD OF ASSAY
Transfer about 400 mg of the dried sample, accurately weighed, into a 250-ml flask, add 20 ml of water and 40 ml of ethanol, cover with a watch glass, heat gently on a steam bath until dissolved and cool. Add 5 drops of phenolphthalein TS and titrate with 0.1 N sodium hydroxide to the first pink colour that persists for 15 sec. Perform a blank determination and make any necessary corrections. Each ml of 0.1 N sodium hydroxide is equivalent to 40.86 mg of C 24 H 40 O 5 .

Identification

Solubility (Vol. 4)
Very slightly soluble in water, soluble in ethanol
Melting range (Vol. 4)
197 - 202 o
Colour reactions
To 1 ml of a 0.02% solution of cholic acid in 50% acetic acid, add 1 ml of a 1% solution of furfural in water, 6 ml water and 5 ml concentrated sulfuric acid. The mixture turns rose and then violet within 5 min. (The same colour is produced by desoxycholic acid). To about 10 mg of cholic acid add 2 drops of benzaldehyde and 3 drops of 75% sulfuric acid and heat at 50 o for 5 min. Then add about 10 ml of glacial acetic acid; a brown colour is produced (desoxycholic acid gives a green colour).

Purity criteria

Loss on drying (Vol. 4)
Not more than 0.5% (140 o , 4 h, pressure less than 5 mm of mercury)
Specific rotation (Vol. 4)
[alpha] 25, D: Not less than + 37 o (2% (w/v) solution in ethanol)
Sulfated ash (Vol. 4)
Not more than 0.1% Test 2 g of the sample
Lead (Vol. 4)
Not more than 2 mg/kg Determine using an atomic absorption technique appropriate to the specified level. The selection of sample size and method of sample preparation may be based on the principles of the method described in Volume 4, “Instrumental Methods.”

Common questions

Assembled from what is above, in the words the questions are usually asked in. Each answer is the same statement the relevant section makes, with the same citations behind it.

What is INS 1000?
INS 1000 is Cholic acid. Cholic acid is a bile acid, used in food as an emulsifier. JECFA's specification describes colourless plates or a white crystalline powder of not less than 98% purity.
What is Cholic acid used for in food?
It is listed as an acidity regulator — the role the US assigns it. This page shows every function each regulator records, with the citation for each.
Is Cholic acid vegan?
No. Cholic acid is animal-derived, and is not vegetarian either. This page says what it is made from.
